抗肿瘤药物在体液中的浓度测定对于改进化疗方法及研究新的药物运载体系均有重要意义。5-氟脲嘧啶(5-FU)测定方法甚多,放射性标记法与抑菌法灵敏度高、干扰小但测定结果易受代谢影响,且缺乏专属性;分光光度法、薄层层析和气液层析法操作方便,但灵敏度较差,速度较慢。作者在De Leenheer等报道的离子对分光光度法基础上加以改进,新方法可快速灵敏地测定生物样品中的5-FU。
The determination of anticancer drug concentrations in body fluids is of great importance to improve the chemotherapy and to study new drug delivery systems. There are many methods for the determination of 5-fluorouracil (5-FU), radiolabeling and antibacterial methods have high sensitivity, small interference but the measurement results are susceptible to metabolism, and the lack of specificity; spectrophotometry, thin layer chromatography and gas-liquid Chromatography easy to operate, but the sensitivity is poor, slower. The authors improve upon the ion-pair spectrophotometry reported by De Leenheer et al. The new method allows rapid and sensitive determination of 5-FU in biological samples.
