目的研究沙利度胺对人宫颈癌Hela细胞增殖及MMP-2、MMP-9的影响。方法培养人宫颈癌Hela细胞并分为不同浓度沙利度胺(B组、C组、D组),分别为100 mg/L、200 mg/L、400 mg/L的沙利度胺和空白对照组(A组),探讨沙利度胺对宫颈癌细胞增殖、凋亡及MMP-2、MMP-9的影响。结果 D组药物作用后12 h、24 h、36 h、48 h时,细胞的增殖率低于A组、B组、C组,B组、C组、D组Hela细胞增值率依次下降,差异有统计学意义(P<0.05)。D组药物作用后12 h、24 h、36 h、48 h时,细胞的凋亡率高于A组、B组、C组,B组、C组、D组Hela细胞凋亡率依次升高。D组患者MMP-2、MMP-9、TIMP中mRNA表达水平低于A组、B组、C组,B组、C组、D组MMP-2、MM-9、TIMP中mRNA浓度依次降低,差异均有统计学意义(P<0.05)。结论沙利度胺可以抑制Hela细胞增殖及MMP-2、MMP-9分泌。
Objective To investigate the effects of thalidomide on the proliferation, MMP-2 and MMP-9 in human cervical carcinoma Hela cells. Methods Human cervical cancer Hela cells were cultured and divided into different concentrations of thalidomide (group B, group C, group D), thalidomide of 100 mg / L, 200 mg / L, 400 mg / L and blank Control group (group A) to investigate the effects of thalidomide on proliferation, apoptosis, MMP-2, MMP-9 in cervical cancer cells. Results The proliferative rate of Hela cells in group B, group C, group B, group C and group D was decreased at 12 h, 24 h, 36 h and 48 h after treatment with D group, There was statistical significance (P <0.05). The apoptotic rates of Hela cells in group B, C, B, C and D were higher than those in group A at 12 h, 24 h, 36 h and 48 h . The mRNA levels of MMP-2, MMP-9 and TIMP in group D were lower than those in group A, B, C, B, C and D, The differences were statistically significant (P <0.05). Conclusion Thalidomide can inhibit Hela cell proliferation and MMP-2, MMP-9 secretion.
