目的和方法 :采用Langendorff大鼠离体心脏灌流技术 ,制备心肌缺氧 /复氧损伤模型 ,分别在缺氧前及缺氧后 ,用复方丹参滴丸及消心痛对心肌给予保护 ,利用高效液相色谱仪 (HPLC)测定离体大鼠心肌组织中的高能磷酸化合物的含量变化 ,利用H - 6 0 0透射电子显微镜对心肌细胞超微结构进行观察。本实验共分 6组 :正常对照组 ;单纯缺氧 /复氧组 ;复方丹参滴丸前、后保护组 ;消心痛前、后保护组。结果 :①单纯缺氧 /复氧组心肌组织中AMP、ADP、ATP及AN含量均明显低于正常对照组 (P <0 0 1)。②复方丹参滴丸前、后保护组大鼠心肌组织内的AMP、ADP、ATP、AN含量均高于单纯缺氧 /复氧组 (P <0 0 1) ,也相应高于消心痛前、后保护组 (P <0 0 1) ,两组的ATP、AN含量均接近正常水平 (P >0 0 5 )。③消心痛前、后保护组的AMP、ADP、ATP、AN含量均低于正常对照组 (P<0 0 1)。电镜结果也证明复方丹参滴丸对缺氧 /复氧心肌细胞超微结构具有明显的保护作用。结论 :复方丹参滴丸无论在缺氧前预灌注时及缺氧后再灌注时给予 ,均能通过提高心肌组织中高能磷酸化合物的含量 ,保护心肌细胞超微结构来保护心肌 ,其保护效果优于消心痛。
OBJECTIVE AND METHOD: To prepare myocardial hypoxia/reoxygenation injury model using Langendorff rat isolated heart perfusion technique. Before and after hypoxia, protect myocardium with compound Danshen Dripping Pills and isosorbide dinitrate. The content of high-energy phosphate compounds in myocardial tissue of isolated rat was measured by phase chromatography (HPLC). The ultrastructure of myocardial cells was observed by transmission electron microscope (H-600). The experiment was divided into 6 groups: normal control group; simple anoxia / reoxygenation group; compound Danshen dripping pill before and after the protection group; isosorbide dinitrate before and after the protection group. Results :1 The contents of AMP, ADP, ATP and AN in myocardial tissue were significantly lower than those in normal control group (P <0 01). 2The contents of AMP, ADP, ATP, and AN in the myocardial tissue of the pre- and post-protective groups of Compound Danshen Dripping Pills were higher than those in the hypoxia/reoxygenation group alone (P < 0.01). In the post-protection group (P < 0 01), the ATP and AN contents in both groups were close to normal levels (P > 0 05). 3 The contents of AMP, ADP, ATP, and AN in the pre- and post-cardiotonic protection group were lower than those in the normal control group (P<0 01). Electron microscopy results also proved that Compound Danshen Dripping Pills had a significant protective effect on the ultrastructure of myocardial cells in hypoxia/reoxygenation. CONCLUSION: Compound Danshen Dripping Pills can be used to protect myocardium by increasing the content of high-energy phosphate compounds in myocardium and preserving the ultrastructure of myocardium, both in pre-perfusion before hypoxia and after hypoxia. Eliminating heartache.
