大孔吸附树脂分离纯化玫瑰花中原花青素的研究

目的:研究大孔吸附树脂分离纯化玫瑰花中原花青素的最优工艺条件。方法:以静态饱和吸附量和解析率为指标,对五种大孔吸附树脂(D101、D1300、NKA、AB-8、NKA-Ⅱ)进行筛选,并以原花青素纯度为指标,采用正交试验法,分别考察了上样浓度、pH、洗脱液浓度和洗脱流速对纯化效果的影响。结果:D101型树脂综合性能最佳,适合纯化玫瑰花中原花青素,最优工艺条件为:上样浓度为1.25 mg/ml,上样pH为2,洗脱液为70%乙醇水溶液,洗脱流速为2 ml/min。结论:本研究可为分离纯化玫瑰花中原花青素提供方法。 Objective: To study the optimum conditions for separation and purification of proanthocyanidins from rose flowers using macroporous adsorption resin. Methods: Five kinds of macroporous adsorption resins (D101, D1300, NKA, AB-8, and NKA-II) were screened with static saturation adsorption capacity and resolution, and the purity of procyanidins was used as an index. Orthogonal test was used. The effects of sample concentration, pH, eluent concentration, and elution flow rate on purification were investigated. RESULTS: The D101 resin had the best overall performance and was suitable for the purification of proanthocyanidins from rose flowers. The optimum conditions were as follows: the sample concentration was 1.25 mg/ml, the sample pH was 2 and the eluent was 70% ethanol solution. It is 2 ml/min. Conclusion: This study can provide methods for the isolation and purification of proanthocyanidins from rose flowers.
