目的观察烧伤膏对重度烧伤模型大鼠早期血浆白介素(inter leukin,IL)-1、肿瘤坏死因子(tumor necrosis foutor,TNF)-α、氧自由基表达的影响。方法在2015年4-6月随机将40只Wistar大鼠分为模型组、烧伤膏治疗组、阳性对照组、正常对照组。利用凝固汽油涂抹皮肤燃烧制备的重度烧伤模型,分别涂抹生理盐水、烧伤膏、百多邦、凡士林软膏。在造模后6、12、24、48 h眼眶取血,通过ELISA方法检测各组大鼠血清中IL-1、TNF-α、通过比色法检测血清超氧化物歧化酶(superoxide disrnutase orgotein,SOD)、丙二醛(malonic dialdehyde,MDA)的表达。同时观察7 d后大鼠存活情况。采用方差分析比较组间差异,P<0.05为差异有统计学意义。结果烧伤膏治疗组与阳性对照组和模型组死亡率分别为30%、70%、90%,烧伤膏组死亡率明显降低,对比差异有统计学意义(P<0.05),烧伤膏组与模型组相比,造模后6、12、24、48 h血清IL-1、TNF-α、MDA的表达明显减少,SOD表达明显增多,对比差异均有统计学意义(均P<0.05)。结论中药烧伤膏可明显减少炎症介质和氧自由基的产生,有利于减轻烧伤后组织损伤和促进创面的愈合。
Objective To observe the effect of Burn Ointment on the expression of interleukin (IL) -1, tumor necrosis factor fator (TNF) -α and oxygen free radicals in severe burn rats. Methods From April to June 2015, 40 Wistar rats were randomly divided into model group, burn cream treatment group, positive control group and normal control group. The use of coagulation peels coated skin burning prepared severe burn model, respectively, coated with saline, Burn ointment, Baidu Bang, Vaseline ointment. Serum IL-1 and TNF-α were detected by ELISA method at 6, 12, 24, 48 h after model establishment. Serum superoxide dismutase SOD) and malonic dialdehyde (MDA). The survival of rats was also observed after 7 days. An analysis of variance was used to compare the differences between groups, P <0.05 was considered statistically significant. Results The mortality rates of burn ointment group and positive control group and model group were 30%, 70% and 90%, respectively. The mortality of burn ointment group was significantly lower than that of the control group (P <0.05) Compared with the control group, the levels of IL-1, TNF-α and MDA decreased significantly at 6, 12, 24 and 48 h after modeling, and the expression of SOD increased significantly. The differences were statistically significant (all P <0.05). Conclusion Chinese medicine burn ointment can significantly reduce the production of inflammatory mediators and oxygen free radicals, is conducive to reducing tissue damage after burn and promote wound healing.
