目的研究南宁市某医院散发性腹泻病例中检出的GII.12型诺如病毒(Norovirus,NV)的基因特征。方法应用荧光定量逆转录-聚合酶链反应,对2007年1月~2008年12月,南宁市某医院门诊腹泻病例的粪便标本进行基因II组(Genogroup Ⅱ,GII)NV核糖核酸检测,阳性标本进行核苷酸序列测定,并构建系统进化树进行分析。结果在检测的696份粪便中,GII组NV阳性180份,阳性率为25.86%。随机挑取30株GII组NV进行核苷酸序列分析,有1株[Nanning(NN,南宁)07194]与GII.12型参考株核苷酸序列同源性达到95.7%~99.1%,在系统进化树中与GII.12型病毒株属同一分支。结论南宁市散发性腹泻病例中存在GII.12型NV感染。
Objective To study the genetic characteristics of Norovirus (GV) type IIII in sporadic diarrhea cases in a hospital in Nanning City. Methods Fluorescent quantitative reverse transcriptase - polymerase chain reaction (RT - PCR) was used to detect Genogroup Ⅱ (GII) NV RNA in stool specimens of outpatients with diarrhea from January 2007 to December 2008 in Nanning City. Positive samples The nucleotide sequence was determined and phylogenetic tree was constructed for analysis. Results Among the 696 fecal samples tested, NV in the GII group was 180 positive with a positive rate of 25.86%. A total of 30 strains of GII were randomly selected for nucleotide sequence analysis. One of them (Nanning (NN) 07194] shared 95.7% -99.1% nucleotide sequence homology with the reference strain of GII.12, The same branch of the phylogenetic tree as the GII.12 strain. Conclusion There is GII.12 NV infection in sporadic diarrhea cases in Nanning.
