目的了解肝癌中整合素的表达概况,探寻在肝癌及正常肝组织中的表达差异。方法以肝癌及癌旁正常肝组织的总 RNA 反转录合成含有α~(32)P dATP 的 cDNA 为探针,与 Atlas 微阵列表达分析膜进行差异杂交,并应用半定量 RT-PCR 验证。结果放射自显影结果经 AtlasImage~(TM)软件分析显示:在所分析的588种已知基因中,整合素相关的基因共有17个,其中 integrin alpha8、betal、beta7、beta8等4个在肝癌组织中表达上调。结论通过 Atlas 微阵列系统分析发现的这些基因的表达改变组成了一个肝癌特异的基因表达谱,是人类首次全面系统地研究肝癌的基因表达改变,整合素基因的差异表达为研究肝癌的侵袭、粘连和转移机制提供了有益的线索。
Objective To understand the expression of integrin in hepatocellular carcinoma (HCC) and explore the differences in expression between hepatocellular carcinoma and normal liver tissue. METHODS: Reverse transcription of total RNA from hepatocellular carcinoma and adjacent normal liver tissues was performed to synthesize α-(32)P dATP-containing cDNA as a probe. Differential hybridization was performed with Atlas microarray expression analysis membrane and semi-quantitative RT-PCR was used for validation. Results The results of autoradiography by AtlasImageTM software showed that there were 17 integrin-related genes among the 588 known genes, of which integrin alpha8, betal, beta7, and beta8 were found in 4 The increase in the expression. Conclusion The expression changes of these genes discovered by Atlas microarray analysis constitute a liver cancer-specific gene expression profile. This is the first comprehensive and systematic study of the gene expression changes of liver cancer in humans. The differential expression of integrin genes is used to study the invasion and adhesion of liver cancer. The transfer mechanism provides useful clues.
