目的:对淡黄香茶菜Isodon flavida地下部分正丁醇部位的化学成分及生物活性进行研究。方法:采用硅胶、羟丙基葡聚糖凝胶(Sephadex LH-20),MCI树脂等多种柱色谱对样品进行分离纯化,运用理化性质和现代波谱技术对单体成分进行结构鉴定;同时,运用1,1-二苯基-2-三硝基苯肼(DPPH)法,β-羟高铁血红素形成抑制法等对样品的抗氧化、抗疟活性进行了初步的研究。结果:从淡黄香茶菜地下部分70%丙酮提取物的正丁醇部位分离得到9个化合物,分别鉴定为2,6-二甲氧基苯醌(1),迷迭香酸甲酯(2),2α-羟基齐墩果酸(3),1-甲氧基-α-D-葡萄糖(4),1-甲氧基-β-D-葡萄糖(5),蔗糖(6),腺花香茶菜苷C(7),5α,6β-二羟基胡萝卜苷(8),(2’R)-1-O-甘油基-6-O-α-D-吡喃半乳糖基-β-D-吡喃半乳糖苷(9);生物活性检测结果显示,化合物2对DPPH有一定的清除作用,同时对β-羟高铁血红素形成也表现出抑制作用,半抑制浓度(IC50)分别为(124.2±2.7),(163.9±7.5)mg·L~(-1)。结论:9个化合物均为首次从该植物中分离得到,化合物2显示出一定的抗疟及抗氧化活性,值得进一步深入研究。本研究为淡黄香茶菜的开发利用提供了一定的实验依据。
Objective: To study the chemical constituents and bioactivities of n-butanol in underground part of Isodon flavida. Methods: The samples were separated and purified by silica gel, Sephadex LH-20, MCI resin and other column chromatographies. The structures of the monomers were identified by physicochemical properties and modern spectroscopic techniques. At the same time, The anti-oxidative and anti-malarial activities of the samples were studied by using 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH) method and β -Hemetobate inhibition method. Results: Nine compounds were isolated from the n-butanol fraction of 70% acetone extract from the underground part of the yellow tea plant Chakra, and were identified as 2,6-dimethoxybenzoquinone (1), methyl rosinate (3), 1-methoxy-α-D-glucose (4), 1-methoxy-β-D-glucose (5), sucrose (8), (2’R) -1-O-glycero-6-O-α-D-galactopyranosyl-β-D- The results of bioassay showed that compound 2 had certain scavenging effects on DPPH and inhibited the formation of β -Hemetabolites. IC50 values were (124.2 ± 2.7), (163.9 ± 7.5) mg · L -1. CONCLUSION: Nine compounds were isolated from this plant for the first time. Compound 2 showed some anti-malarial and anti-oxidative activity, which deserved further study. This study provides a certain experimental basis for the development and utilization of light yellow tea.
