建立比格犬血浆中低浓度盐酸二甲双胍高效液相色谱测定方法,用于研究盐酸二甲双胍在消化道各部位的吸收情况。采用高效液相色谱法,以dikma Diamonsil C18(150 mm×4.6 mm,5μm)柱为分析柱,乙腈-10.0 mmol/L磷酸二氢钠水溶液(含7.5 mmol/L十二烷基磺酸钠,pH为5.5)(32:68)为流动相,柱温:30℃,检测波长:235 nm,流速:1.0 mL/min,内标法检测。结果表明,盐酸二甲双胍与内标分离良好,回归方程Y=0.6868X+(-0.0222)r=0.9995,线性范围0.05~4.0μg/mL。方法回收率在99.78%~107.90%,批内RSD≤3.85%,批间RSD≤5.63%。
To establish a beagle dog plasma low concentration of metformin hydrochloride HPLC method for the study of metformin hydrochloride absorption in various parts of the digestive tract. High performance liquid chromatography (HPLC) was performed on a dikma Diamonsil C18 (150 mm × 4.6 mm, 5 μm) column with acetonitrile-10.0 mmol / L aqueous sodium dihydrogen phosphate solution containing 7.5 mmol / L sodium dodecyl sulfate, (pH = 5.5) (32:68) as mobile phase. The column temperature was 30 ℃, the detection wavelength was 235 nm, the flow rate was 1.0 mL / min and the internal standard method was used. The results showed that metformin hydrochloride was separated from the internal standard well. The regression equation was 0.6868X + (- 0.0222) r = 0.9995 with a linear range of 0.05 ~ 4.0μg / mL. The recoveries of the method ranged from 99.78% to 107.90%. The intra-assay RSD was less than 3.85%. Inter-assay RSD was less than or equal to 5.63%.
