目的:建立去卵巢大鼠骨质疏松症动物模型,检测造模成功后不同时期大鼠胫骨OPG/RANKLmRNA表达量及血清雌二醇水平,分析OPG/RANKLmRNA表达变化规律。方法:选取3月龄雌性SD大鼠60只随机分为2组即去卵巢组与对照组,每组30只。去卵巢组去除双侧卵巢,对照组仅去除腹腔少许脂肪。分别在造模12周后0个月、6个月、12个月每组随机处死10只,检测不同时期大鼠胫骨OPG/RANKLmRNA表达量及血清雌二醇水平。采用SPSS 17.0软件进行统计,分析数据,得出结论。结果:与对照组相比,去卵巢组胫骨OPGmRNA表达量、血清雌二醇浓度在各时间点上均显著降低(P<0.05),RANKLmRNA表达量则显著增高(P<0.05);去卵巢组0个月与6个月、6个月与12个月比较,胫骨OPGmRNA表达量及血清雌二醇浓度下降,差异具有统计学意义(P<0.05),RANKLmRNA表达量增高,差异具有统计学意义(P<0.05)。结论:造模成功后12个月内,去卵巢大鼠胫骨骨组织OPGmRNA表达量持续减少,血清雌二醇浓度均持续降低,RANKLmRNA持续高表达,OPG/RANKL比值缩小。
OBJECTIVE: To establish an animal model of osteoporosis in ovariectomized rats. The expression level of OPG / RANKL mRNA and the level of serum estradiol in rat tibia at different periods after successful modeling were determined. The changes of OPG / RANKL mRNA expression were analyzed. Methods: Sixty female 3-month-old female Sprague-Dawley rats were randomly divided into 2 groups: ovariectomized group and control group, with 30 in each group. Ovary group to remove both ovaries, the control group only remove a little fat in the abdominal cavity. Ten rabbits in each group were randomly sacrificed at 0, 6 and 12 months after 12 weeks of modeling. The expression of OPG / RANKL mRNA and the level of serum estradiol in the tibia of rats at different periods were measured. Using SPSS 17.0 software for statistics, analysis of data, draw conclusions. Results: Compared with the control group, the expression of OPG mRNA and serum estradiol in the tibia of the ovariectomized group were significantly decreased (P <0.05) and the expression of RANKL mRNA was significantly increased in the ovariectomized group (P <0.05) The expression of OPG mRNA and serum estradiol in tibia at 0, 6 months, 6 months and 12 months decreased significantly (P <0.05), and the expression of RANKL mRNA increased, the difference was statistically significant (P <0.05). CONCLUSION: Within 12 months after the success of modeling, the expression of OPG mRNA in the tibial bone of ovariectomized rats continuously decreases, the serum estradiol concentration continuously decreases, the expression of RANKL mRNA continues to be high, and the ratio of OPG / RANKL is reduced.
