目的:探讨肾消通络方对糖尿病肾病大鼠TGF-β1/Smad信号传导通路的影响。方法:将实验动物分为正常组、模型组、厄贝沙坦组及中药组4组,免疫组化法测定肾组织TGF-β1蛋白表达,RT-PCR法测定TGF-β1mRNA表达,Western blot法测定Smad2/3及Smad7的表达。结果:中药组和厄贝沙坦组TGF-β1蛋白的表达水平较模型组有所降低,TGF-β1 mRNA水平明显低于模型组,Smad2/3水平明显低于模型组,Smad7水平明显高于模型组。结论:肾消通络方可抑制TGF-β1mRNA及其蛋白的表达,同时抑制Smad2/3表达而提高Smad7的表达。
Objective: To investigate the effect of Shenxuetongluo prescription on TGF-β1 / Smad signal transduction pathway in diabetic nephropathy rats. Methods: The experimental animals were divided into four groups: normal group, model group, irbesartan group and traditional Chinese medicine group. The expression of TGF-β1 protein in renal tissue was detected by immunohistochemical method. The expression of TGF-β1 mRNA was detected by RT- The expression of Smad2 / 3 and Smad7 was determined. Results: The expression of TGF-β1 in Chinese medicine group and irbesartan group was lower than that in model group, the level of TGF-β1 mRNA was significantly lower than model group, the level of Smad2 / 3 was significantly lower than that of model group, the level of Smad7 was significantly higher than that of model group Model group. Conclusion: Shenxiao Tongluo can inhibit the expression of TGF-β1 mRNA and protein, inhibit the expression of Smad2 / 3 and increase the expression of Smad7.
