目的:探讨不同程度的慢性酒精中毒对大鼠耳蜗神经肽VIP(血管活性肠肽)及其受体表达的影响及其机制。方法:30只SD大鼠随机分为3组进行造模:轻度酒精中毒组、重度中毒组及对照组。采用RT-PCR分别检测3组大鼠耳蜗VIP的表达,采用Western blotting检测大鼠耳蜗VIP受体蛋白的表达,采用免疫组织化学的方法定位及定量检测大鼠耳蜗VIP的表达。结果:慢性酒精中毒组的大鼠耳蜗VIP及其受体的表达下调,并且重度慢性酒精中毒的大鼠耳蜗VIP及其受体的表达低于轻度组,差异有统计学意义(P<0.05)。慢性酒精中毒组的大鼠耳蜗VIP及其受体与对照组在神经节细胞的表达差异无统计学意义(P>0.05)。结论:VIP及其受体在慢性酒精中毒大鼠的听觉系统功能中发挥着重要的作用。慢性酒精中毒可引起听觉系统的一种肽类激素分泌不平衡,最终导致听力损失。
Objective: To investigate the effects of chronic alcoholism on the expression of VIP (vasoactive intestinal peptide) and its receptor in rat cochlea and its mechanism. Methods: Thirty SD rats were randomly divided into three groups: mild alcoholism group, severe poisoning group and control group. The expression of VIP in the rat cochlea was detected by RT-PCR. The expression of VIP receptor protein in the rat cochleas was detected by Western blotting. The expression of VIP in the rat cochleas was detected by immunohistochemistry. Results: The expression of VIP and its receptor in rat ’s cochlea was decreased in chronic alcoholism group, and the expression of VIP and its receptor in rat with severe chronic alcoholism was lower than that in mild group (P <0.05 ). There were no significant differences in the expression of VIP and its receptor in the rat with chronic alcoholism and the control group in ganglion cells (P> 0.05). Conclusion: VIP and its receptors play an important role in the auditory system function of chronic alcoholism rats. Chronic alcoholism can cause an imbalance in the secretion of a peptide hormone in the auditory system, eventually leading to hearing loss.
