以红花檵木为研究对象,初步探索其组织培养技术。试验选取健壮的半木质化茎段作为外植体,通过消毒效果和芽萌发率比较,进行外植体不同消毒方法和启动培养基的筛选。试验结果表明:采用0.1%HgCl2消毒2次(4分钟/次),再用50 mmol/L CaCl2浸洗3次(5分钟/次)的消毒方式最有利于外植体启动,可获得大量有效的无菌试验材料;选择MS+6-BA(1.00 mg/L)+IBA(0.02 mg/L)+VC(5.00mg/L)做启动培养基较为合适,启动率达83.33%,死亡率仅为16.67%。
Safflower as the research object, preliminary exploration of its tissue culture technology. Selection of robust semi-lignified stem segments as explants, through the disinfection effect and bud germination rate compared to different explants of different disinfection methods and start-up medium screening. The results showed that disinfection with 0.1% HgCl2 twice (4 minutes / time) and then with 50 mmol / L CaCl2 for 3 times (5 minutes / time) was the most effective for explants to start, and a lot of effective (1.0 mg / L) + IBA (0.02 mg / L) + VC (5.00 mg / L) as starter medium, with a starting rate of 83.33% and a mortality of only 16.67%.
