目的 对大鼠心、脑、骨骼肌毛细血管染色方法进行比较。方法 采用Mg2 + ATPase、碱性磷酸酶、肌球蛋白 ATPase等酶组化法及墨汁灌注法和Ⅷ 因子相关抗原免疫组化法对毛细血管进行染色。结果 心脏和脑组织毛细血管用碱性磷酸酶染色 ,而骨骼肌组织毛细血管用肌球蛋白 ATPase染色效果最好 ;Ⅷ 因子相关抗原免疫组化法用于脑组织毛细血管显示次之 ;Mg2 + ATPase法在心脏、脑、骨骼肌定位效果较差 ;墨汁灌注法只要方法掌握得当 ,各部位毛细血管显示均较好。结论 对毛细血管染色 ,不同器官和组织应采用不同的方法。在心脏和脑用碱性磷酸酶组化法显示较好 ,而在骨骼肌用肌球蛋白 ATPase酶组化法较好。
Objective To compare the capillary staining of rat heart, brain and skeletal muscle. Methods Capillary vessels were stained with Mg 2+ -ATPase, alkaline phosphatase, myosin ATPase, and ink perfusion and factor Ⅷ-related antigen immunohistochemistry. Results The capillary of heart and brain tissue was stained with alkaline phosphatase, while the skeletal muscle capillary was best stained with myosin ATPase. The Ⅷ factor-related antigen immunohistochemical method was used to detect the capillary of brain tissue followed by Mg2 + ATPase method in the heart, brain, skeletal muscle positioning less effective; ink perfusion method as long as the method is properly controlled, all parts of the capillaries showed better. Conclusions For capillary staining, different organs and tissues should be treated differently. Alkaline phosphatase in the heart and brain showed good histochemistry, while skeletal muscle with myosin ATPase enzyme histochemistry is better.
