B细胞活化因子(BAFF)的异常表达与骨髓瘤、淋巴瘤的发病相关,为探索常见炎性细胞因子是否能影响人骨髓白血病细胞HL-60中BAFF的表达,以细胞因子IFN-γ、IL-10和IL-4刺激体外培养的HL-60细胞1~3 d后,采用流式细胞术、ELISA和荧光定量RT-PCR方法检测BAFF的核酸和蛋白表达含量的变化。结果显示,细胞因子IFN-γ、IL-10干预后,HL-60细胞表面BAFF蛋白表达水平、细胞上清液中BAFF蛋白含量、细胞内BAFF mRNA的含量均能明显升高(P<0.05),其中在INF-γ、IL-10干预的第1天,HL-60细胞内BAFF mRNA的含量为最高;在干预的第2天,HL-60细胞的上清液中BAFF蛋白含量及细胞表面BAFF蛋白表达水平均达最大值;而IL-4干预HL-60细胞后,仅对细胞上清液中BAFF蛋白水平稍有降低(P<0.05),细胞表面BAFF蛋白表达水平和细胞内BAFF mRNA的含量均无显著性差异。研究提示,INF-γ、IL-10可能是通过促进HL-60细胞对BAFF核酸的表达来上调BAFF蛋白的合成与分泌,而IL-4则仅能一定程度的降低HL-60细胞对BAFF蛋白的分泌,此研究为探讨骨髓白血病发病过程中,BAFF异常表达的调控机制提供了依据。
Abnormal expression of B-cell activating factor (BAFF) is associated with the pathogenesis of myeloma and lymphoma. To explore whether common inflammatory cytokines can affect the expression of BAFF in human myeloid leukemia HL-60 cells, The expression of BAFF mRNA and protein in HL-60 cells were detected by flow cytometry, ELISA and real-time quantitative RT-PCR after stimulated with IL-10 and IL-4 for 1-3 days respectively. The results showed that BAFF protein expression, BAFF protein content in cell supernatant, and BAFF mRNA content in HL-60 cells were significantly increased after IFN-γ and IL-10 intervention (P <0.05) , In which the content of BAFF mRNA in HL-60 cells was the highest on the first day of INF-γ and IL-10 intervention; on the second day of intervention, the content of BAFF in HL-60 cells supernatant and the cell surface BAFF protein expression levels reached the maximum; IL-4 intervention HL-60 cells, only the cell supernatant BAFF protein levels decreased slightly (P <0.05), cell surface BAFF protein levels and intracellular BAFF mRNA No significant difference in content. The results suggest that IL-4 may up-regulate the synthesis and secretion of BAFF protein by promoting the expression of BAFF in HL-60 cells, but IL-4 may only reduce the effect of BAFF protein This study provides a basis for exploring the regulatory mechanism of abnormal expression of BAFF in the pathogenesis of myeloid leukemia.
