用新生大鼠胰岛离体培养方法,观察了五肽促胃液素(G-5)对白细胞介素-1β(IL-1β)致胰岛B细胞基础分泌抑制的影响。结果发现:①IL-1β能剂量依赖性抑制B细胞的分泌功能,IL-1β作用组基础胰岛素分泌量明显低于对照组;②C-5能对抗IL-1β对B细胞功能的抑制,与单纯IL-1β作用组相比,加用G-5能使基础胰岛素分泌量明显增加;③向IL-1β作用后的胰岛治疗性地给予G-5,能使抑制的B细胞功能得到部分恢复,作用呈时间和剂量依赖性。提示:G-5可能具有对抗IL-1β对胰岛B细胞毒性损伤的作用。
The effect of pentagastrin (G-5) on the inhibition of basal secretion of islet B cells induced by interleukin-1β (IL-1β) was observed in cultured rat islets isolated from neonatal rats. The results showed that: (1) IL-1β can inhibit the secretion of B cells in a dose-dependent manner, and the basal insulin secretion of IL-1β group was significantly lower than that of control group; ②C-5 could antagonize the inhibitory effect of IL-1β on B cells, G-5 significantly increased basal insulin secretion compared with that of G-5; G-5 was administered to islets treated with IL-1β, which partially restored the function of inhibited B cells Time-and dose-dependent. It is suggested that G-5 may have the effect of antagonizing the cytotoxic damage of IL-1β to pancreatic islet B cells.
