树突状细胞(DC)是免疫系统中一类强有力的抗原提呈细胞(APC),在始动机体免疫反应中起至关重要的作用。近年来,人们极大地关注DC用于免疫治疗的前景。然而,DC在人外周血中含量甚微(约占外周血单个核细胞的0.1%),给研究带来相当困难。本研究联合应用rhGM-CSF,rhIL-4和rhTNF-α从人外周血中体外培养富集获得高纯度DC群,通过DC的特有形态鉴别以及应用X-11单克隆抗体(抗人血DC表面特异抗原决定簇的单克隆抗体),借助于间接免疫荧光试验(IFA)及透射电镜观察,初步描述了所获取的DC群的特征。这些结果为进一步观察人外周血DC的超微结构和细胞化学特征及其临床研究DC功能提供了基础。
Dendritic cells (DCs) are a class of potent antigen presenting cells (APCs) in the immune system that play a crucial role in initiating immune responses in the body. In recent years, great attention has been paid to the prospect of DC for immunotherapy. However, DC is very low in human peripheral blood (about 0.1% of peripheral blood mononuclear cells), which brings quite difficult research. In this study, a combination of rhGM-CSF, rhIL-4 and rhTNF-α was cultured and enriched from human peripheral blood in vitro to obtain high-purity DCs.
