目的观察红背叶根4种不同提取物对HepG 2.2.15细胞分泌HBsAg和HBeAg的影响,探讨红背叶根不同提取物体外抗HBV作用。方法通过四甲基偶氮唑盐(MTT)法观察红背叶根水提取物(WE)、石油醚提取取物(EE)及正丁醇提取物(BE)对HepG 2.2.15细胞的影响,采用ELISA法检测分析红背叶根4种提取物对HepG 2.2.15细胞分泌HBsAg和HBeAg的抑制作用。结果红背叶根正丁醇提取物在无毒浓度下对HepG2.2.15分泌的HBsAg、HBeAg的抑制率分别达65.2%、94.4%,治疗指数分别为>9.8和>67.1;红背叶根乙酸乙酯提取物对HBeAg抑制率达53.4%,治疗指数>2,而对HBsAg的无抑制作用;其他两种提取物对HBsAg、HBeAg则无抑制效果。结论红背叶根乙酸乙酯提取物、正丁醇提取物均有抗HBV作用,正丁醇提取物效果比乙酸乙酯效果更好,具有潜在的抗HBV开发前景。
Objective To observe the effects of 4 different extracts of red dorsal root on the secretion of HBsAg and HBeAg in HepG 2.2.15 cells and to explore the anti-HBV effect of different extracts from red dwarf roots. Methods The effects of water extract (WE), petroleum ether extract (EE) and n-butanol extract (BE) on HepG 2.2.15 cells were observed by MTT assay. The inhibitory effect of 4 extracts of red dorsal root on the secretion of HBsAg and HBeAg in HepG 2.2.15 cells was detected by ELISA. Results The inhibitory rates of HBsAg and HBeAg secreted by HepG2.2.15 in non-toxic concentrations were 65.2% and 94.4%, respectively, and the therapeutic indexes were> 9.8 and> 67.1 respectively. Ethyl ester extract of HBeAg inhibition rate of 53.4%, treatment index> 2, but no inhibition of HBsAg; the other two extracts on HBsAg, HBeAg no inhibitory effect. Conclusion Ethyl acetate extract and n-butanol extract of red dorsal root root have anti-HBV effects. N-butanol extract is more effective than ethyl acetate and has potential anti-HBV development prospect.
