对白灵菇子实体粗多糖进行分离纯化与纯度鉴定,并应用化学发光法对多糖清除羟自由基(.OH)和超氧阴离子自由基(O2.)的能力进行研究。结果表明,利用Savag法脱法蛋白质、透析法脱法小分子物质以及醇沉后得到的粗提物,经DEAE-Cellulose52和SephadexG-200柱层析后得到纯品;采用UV、琼脂糖凝胶电泳以及SephadexG-200方法鉴定这种物质PNFPⅡ-a是一种纯多糖;而且这种子实体纯多糖具有较强的抗氧化活性,能显著消除O2.和.OH。
Purification and purity identification of crude polysaccharide from Pleurotus nebrodensis fruiting bodies were studied, and the ability of polysaccharide to scavenge hydroxyl radical (.OH) and superoxide anion radical (O2.) Was studied by chemiluminescence. The results showed that the protein was purified by Savag method, and the small molecules separated by dialysis method and the crude extracts obtained by alcohol precipitation were purified by DEAE-Cellulose 52 and SephadexG-200 column chromatography. UV, agarose gel electrophoresis and SephadexG-200 method to identify this substance PNFP Ⅱ-a is a pure polysaccharide; and this fruiting body polysaccharides have strong antioxidant activity, can significantly eliminate O2.
