目的观察促性腺激素释放激素类似物曲普瑞林逆转卵巢恶性肿瘤对顺铂耐药性的效果。方法建立卵巢肿瘤顺铂耐药细胞系模型OVCAR-3/CDDP,采用MTT比色法分别检测单独使用曲谱瑞林、单独使用顺铂、曲谱瑞林和顺铂联用时对OVCAR-3/CDDP细胞的抑制效果,根据血药峰值浓度的不同分成顺铂组、曲谱瑞林组、两药联合组和OVCAR-3组。观察各组的细胞生长情况,分析曲谱瑞林对OVCAR-3/CDDP细胞顺铂耐药性的逆转作用。结果顺铂组、曲谱瑞林组、OVCAR-3组细胞的生长状况相近,细胞在不同时间点的生长抑制率差异无统计学意义(P>0.05);而两药联合组的细胞生长抑制率与其他3组比较,差异有统计学意义(P<0.05)。与顺铂组、曲谱瑞林组相比,顺铂和曲谱瑞林联用组能更显著地削弱OVCAR-3/CDDP细胞的耐药性。结论曲谱瑞林能在一定程度上抑制卵巢肿瘤顺铂耐药细胞的生长,逆转卵巢肿瘤顺铂耐药细胞系的耐药性。
Objective To observe the effects of triptorelin, a gonadotropin-releasing hormone analogue, on reversing the drug resistance of ovarian cancer. Methods OVCAR-3 / CDDP cell line was established in OVCAR-3 / CDDP cell line. MTT assay was used to detect the inhibition of OVCAR-3 / CDDP cells when used alone or combined with cisplatin, The effect was divided into cisplatin group, songruilin group, two drug combination group and OVCAR-3 group according to the peak plasma concentration. The growth of cells in each group was observed, and the reversal effect of CETR on cisplatin resistance in OVCAR-3 / CDDP cells was analyzed. Results The growth of OVCAR-3 cells in Cisplatin group, CTSR group and OVCAR-3 group was similar, and there was no significant difference in the growth inhibition rate of cells at different time points (P> 0.05). However, the cell growth inhibition rate Compared with the other three groups, the difference was statistically significant (P <0.05). Compared with Cisplatin group and CrestRarelin group, cisplatin combined with Cistronicin significantly weakened the drug resistance of OVCAR-3 / CDDP cells. Conclusion Ciprofwelin can inhibit the growth of cisplatin-resistant cells in ovarian tumors to a certain extent and reverse the drug resistance of cisplatin-resistant ovarian cancer cell lines.
