目的在确定天麻中天麻素和巴利森苷是入血成分的基础上,采用反相高效液相色谱法建立了天麻素和巴利森苷的含量测定方法。方法采用Agilent Elipse XDB-C18(4.6 mm×150 mm,5μm)色谱柱,柱温30℃,检测波长270 nm,流动相为甲醇-1%冰醋酸溶液,流速0.8 m L·min-1。结果天麻素和巴利森苷的线性关系良好,具有较好的精密度和稳定性。天麻素的线性范围为10~200μg·m L-1,回归方程为Y=0.0346X-0.0397,r=0.999 8;巴利森苷的线性范围为20~400μg·m L-1,回归方程为Y=0.0236X-0.0016,r=0.999 9;平均回收率分别为98.2%,97.2%。结论该方法具有良好的重复性和回收率,可为天麻药材的质量控制提供参考。
Objective To determine the content of gastrodin and barison glycosides in Gastrodia elata Blume based on the analysis of the components of gastrodin and Ballyllin glycosides into the bloodstream by reversed-phase high performance liquid chromatography. Methods The column of Agilent Elipse XDB-C18 (4.6 mm × 150 mm, 5 μm) was used. The column temperature was 30 ℃ and the detection wavelength was 270 nm. The mobile phase consisted of methanol-1% acetic acid solution and the flow rate was 0.8 m L · min-1. Results Gastrodin and Ballysin glycosides good linear relationship, with good precision and stability. The linear range of gastrodin was 10 ~ 200μg · m L-1, the regression equation was Y = 0.0346X-0.0397, r = 0.999 8, and the linear range of Bareisin was 20-400μg · m L-1. The regression equation was Y = 0.0236X-0.0016, r = 0.999 9; the average recoveries were 98.2% and 97.2% respectively. Conclusion The method has good repeatability and recovery, which can provide a reference for the quality control of Gastrodia elata Blume.
