本文观察了中药钙拮抗剂汉防己甲素对大鼠环孢素A肾毒性(CsA-NT)的防护作用。雄性Wistar大鼠用CsA50mg·kg-1/d灌胃7天导致急性CsA-NT,表现为血清肌酐(Cr)升高34.8%,内生肌酐清除率(Ccr)下降39.4%,组织学检查发现近曲小管大片空泡变性。同时,肾皮质丙二醛(MDA)含量显著升高,超氧化物歧化酶(SOD)活性显著下降。大鼠每日用CsA灌胃前30分钟,给予汉防己甲素10mg/kg腹腔注射,能使上述指标明显改善,组织学损伤也明显减轻。实验结果提示汉防己甲素能从功能和形态上防护大鼠急性CsA-NT。
In this paper, we observed the protective effect of traditional Chinese medicine calcium antagonist tetrandrine on cyclosporine A nephrotoxicity (CsA-NT) in rats. Male Wistar rats were treated with CsA50mg·kg-1/d for 7 days to induce acute CsA-NT with a 34.8% increase in serum creatinine (Cr) and a 39.4% decrease in creatinine clearance (Ccr). Histological examination revealed large vacuolar degeneration of proximal tubules. At the same time, the content of malondialdehyde (MDA) in the kidney cortex was significantly increased, and the activity of superoxide dismutase (SOD) was significantly decreased. Rats were given intraperitoneal injection of tetrandrine 10 mg/kg 30 min before gavage every day with CsA, which can significantly improve the above-mentioned indicators and significantly reduce histological damage. The experimental results suggest that tetrandrine can protect rat acute CsA-NT functionally and morphologically.
