应用一株T淋巴母细胞CEM免疫BALB/c小鼠后,取脾细胞与SP2/O职合,分别筛选出3株分泌抗β_2-微球蛋白(β_2-m)单抗细胞株,它们的反应性均能被纯化的可溶性β_2-m完全抑制,但D_(11)E_3、H_5B_2两种单抗既能与游离的β2m反应,亦能与细胞膜表面HLA-β_2-m复合物反应,它们识别相同或者权相近的抗原决定簇。相反,D_8E_3抗体不与细胞膜表面抗原反应,经抗体竞争实验及加成实验,均证实D_8E_3抗体所识别的抗原决定簇,与D_(11)E_3、H5B2不同。推测在细胞膜表面,D_8E_3抗体所识别的抗原决定簇可能被HLA重链相关区域所覆盖。
After BALB / c mice were immunized with C-T lymphocytes and spleen cells were incubated with SP2 / O serogroups, three strains of McAbs secreting anti-β_2-microglobulin (β_2-m) were screened. The reactivity of the two antibodies could be completely inhibited by the purified soluble β_2-m. However, the two monoclonal antibodies D_ (11) E_3 and H_5B_2 reacted with free β2m and with the HLA-β_2-m complex on the surface of cell membrane, The same or similar antigenic determinants. On the contrary, D_8E_3 antibody did not react with cell surface antigens, and the anti-D_8E_3 antibody recognized the antigenic determinant by the antibody competition experiment and addition test, which was different from D_ (11) E_3 and H5B2. It is speculated that on the cell membrane surface, the epitopes recognized by the D_8E_3 antibody may be covered by HLA heavy chain related regions.
