目的为了阐明汉滩病毒囊膜蛋白及核壳蛋白与免疫保护力的关系。方法采用区带离心及柱层析纯化汉滩病毒LR1株囊膜蛋白GP和核壳蛋白NP,以GP、NP和GP+NP分别免疫家兔,测定其中和抗体和保护力。结果实验显示,GP和GP+NP免疫组家兔血清中和抗体滴度≥110,NP免疫组家兔血清中和抗体≤15;而保护性试验显示,GP、NP、GP+NP免疫组家兔均可保护100ID50同株病毒的攻击。结论上述结果表明,汉滩病毒GP和NP组份均与其保护力有关。
Objective To elucidate the relationship between Hantaan virus envelope protein and nucleocapsid protein and immune protection. Methods The glycoprotein (GP) and nucleocapsid protein (NP) of Hantaan virus LR1 strain were purified by band centrifugation and column chromatography. The rabbits were immunized with GP, NP and GP + NP respectively, and the neutralizing antibody and protective ability were measured. Results The results showed that the titers of neutralizing antibodies in GP and GP + NP rabbits were ≥110, and the serum neutralizing antibodies of NP rabbits were ≤15. Protective tests showed that GP, NP and GP + NP immunized rabbits Rabbits can protect 100ID50 homologous virus attacks. Conclusion The above results indicate that both the GP and NP components of Hantaan virus are related to their protective ability.
