目的探讨乳腺癌组织中血管内皮生长因子(VEGF)-(A、C、D)mRNA表达在乳腺癌发生发展中的作用及其与预后的关系。方法用TaqMan real-time逆转录聚合酶链反应检测61例乳腺癌和29例乳腺良性病变中VEGF-(A、C、D)mRNA的表达,分析其表达与乳腺癌临床病理参数及生存状况间的关系。结果(1)乳腺癌中VEGF-(A、C)mRNA表达量(2.79±1.31、3.33±0.88)高于良性乳腺病变(1.59±1.35、2.76±0.55),差异有统计学意义(P=0.000,P=0.002)。乳腺癌中VEGF-D mRNA表达阳性率(73.77%)高于良性乳腺组织(51.72%,P=0.038),但表达量二者间无差异,P=0.683。(2)单因素和多因素分析均显示VEGF-D与VEGF-C mRNA比值在淋巴结转移阳性组低于阴性组,且与淋巴结转移有关(P单=0.046,P多=0.062)。(3)乳腺癌VEGF-(A、C)mRNA高表达患者无病生存率分别低于低表达组(P=0.030,P=0.044)。结论VEGF-(A、C、D)mRNA表达异常可能与乳腺癌发生发展有关。VEGF—D与VEGF-C mRNA表达比值改变可能与乳腺癌淋巴结转移有关,VEGF-(A、C)是乳腺癌预后重要的影响因子。
Objective To investigate the role of vascular endothelial growth factor (VEGF) - (A, C, D) mRNA expression in the development of breast cancer and its relationship with prognosis in breast cancer. Methods The expression of VEGF- (A, C, D) mRNA in 61 cases of breast cancer and 29 cases of benign breast lesions were detected by TaqMan real-time reverse transcription polymerase chain reaction. The relationship between the expression of VEGF- Relationship. Results (1) The expression of VEGF- (A, C) mRNA in breast cancer was significantly higher than that in benign breast lesions (2.79 ± 1.31,3.33 ± 0.88) (1.59 ± 1.35,2. 76 ± 0.55), the difference was statistically significant (P = 0.000, P = 0.002). The positive rate of VEGF-D mRNA in breast cancer (73.77%) was higher than that in benign breast tissue (51.72%, P = 0.038), but the difference was not significant (P = 0.683). (2) Both univariate and multivariate analysis showed that the ratio of VEGF-D to VEGF-C mRNA in lymph node metastasis positive group was lower than that in negative group (P = 0.046, P = 0.062) . (3) The disease-free survival rates of patients with high expression of VEGF- (A, C) mRNA in breast cancer were lower than those in patients with low expression (P = 0.030, P = 0.044). Conclusion The abnormal expression of VEGF- (A, C, D) mRNA may be related to the development of breast cancer. The change of VEGF-D and VEGF-C mRNA expression may be related to the lymph node metastasis of breast cancer. VEGF- (A, C) is an important factor affecting the prognosis of breast cancer.
