目的 :研究猪囊尾蚴副肌球蛋白 (Ag B)核酸疫苗诱发的免疫应答。 方法 :将 pc DNA3- Ag B核酸疫苗肌注 4~ 6周龄的昆明种小鼠 ,从 2周后开始用 EL ISA方法检测小鼠体内 Ig G和 Ig G2 a的水平。另外将该核酸疫苗注射 C5 7BL / 6小鼠 ,体外培养小鼠脾细胞 ,用刀豆蛋白 A (Con A)和特异性抗原刺激 ,EL ISA试剂盒检测细胞因子 IL - 2和 IL - 4的水平 ,并观察脾淋巴细胞增殖反应。另外 ,将该核酸疫苗肌注瘦型仔猪 ,进行攻击实验 ,计算相对保护率。结果 :注射疫苗后的第 3周起 ,Ig G即为阳性 ,持续升高至第 8周 ;从第 2周起 ,小鼠血清 Ig G2 a为阳性 ,第 4周达到高峰。细胞因子检测表明 ,IL- 2水平远远高于空载体对照组 ,而 IL- 4含量却与空载体对照组无显著差异 ;实验组刺激指数明显高于空载体对照组。副肌球蛋白核酸疫苗对仔猪的相对保护率为 80 %。 结论 :猪囊尾蚴副肌球蛋白核酸疫苗在小鼠动物实验中引发了以 Th1型免疫应答占优势的全面免疫应答。pc DNA3- Ag B核酸疫苗对仔猪具有较好的免疫保护性。
Objective: To study the immune response induced by the vaccine of Cysticercus cellulosae para-myosin (Ag B). Methods: Kunming mice aged 4 to 6 weeks were intramuscularly injected with pcDNA3-Ag B DNA vaccine. The levels of Ig G and Ig G2 a in mice were detected by EL ISA method two weeks later. In addition, the vaccine was injected into C5 7BL / 6 mice and the splenocytes of mice were cultured in vitro. The splenocytes were stimulated with Con A and specific antigen. EL ISA kit was used to detect the cytokines IL - 2 and IL - 4 Level, and observed spleen lymphocyte proliferation reaction. In addition, the nucleic acid vaccine was injected intramuscularly into lean piglets to test the relative protection ratio. Results: Ig G was positive from the third week after vaccination, and continued to rise to the eighth week. From the second week, Ig G2 a was positive in the serum of the mice and peaked at the fourth week. Cytokines showed that the level of IL-2 was much higher than that of empty vector control group, while the content of IL-4 was not significantly different from that of empty vector control group. The stimulation index of experimental group was obviously higher than that of empty vector control group. Paramyosin nucleic acid vaccine relative protection of piglets was 80%. CONCLUSIONS: Cysticercus cellulosae para-myosin nucleic acid vaccine elicits an overall immune response that predominates in a Th1-type immune response in mouse experiments. pc DNA3-Ag B nucleic acid vaccine has good immunoprotection to piglets.
