目的:分析小球藻精油的主要成分,研究其对人结肠癌细胞的生长抑制和凋亡诱导作用机制。方法:GCMS分析小球藻精油的挥发性成分,用不同含量的小球藻精油处理人结肠癌细胞(HT29细胞),采用四甲基偶氮唑蓝(MTT)法检测细胞增殖,流式细胞仪检测细胞凋亡率,分析细胞周期。结果:小球藻精油的主要成分为十六烷(49.69%)、乙酸(17.51%)、乙酸乙酯(10.36%)、甲苯(9.95%)、正十四碳烷(7.96%)、2,4-二甲基环己醇(2.94%)等,其羟基及不饱和双键是抗肿瘤作用的结构基础。与对照组相比,小球藻精油呈剂量依赖性抑制HT29细胞增殖(P<0.01),并引起明显的细胞周期阻滞,将细胞周期阻滞在S(G2/M)期,从而促进细胞凋亡。结论:小球藻精油的挥发性成分和脂肪酸对结肠癌细胞具有增殖抑制效应及凋亡诱导效应,是一种具有发展潜力的抗结肠癌药物。
OBJECTIVE: To analyze the main components of Chlorella essential oil and study its mechanism of growth inhibition and apoptosis induction in human colon cancer cells. Methods: GCMS was used to analyze the volatile components of chlorella essential oil, human colon cancer cells (HT29 cells) were treated with different levels of chlorella essential oil, cell proliferation was detected by MTT assay, flow cytometry Instrument detection of apoptosis rate, analysis of cell cycle. Results: The main components of chlorella essential oil were hexadecane (49.69%), acetic acid (17.51%), ethyl acetate (10.36%), toluene (9.95%), n-tetradecane 4-dimethylcyclohexanol (2.94%) and so on, the hydroxyl and unsaturated double bond is the structural basis of anti-tumor effect. Compared with the control group, the chlorella essential oil inhibited the proliferation of HT29 cells in a dose-dependent manner (P <0.01), and caused significant cell cycle arrest, arresting the cell cycle in S (G2 / M) phase, Apoptosis. Conclusion: The volatile components of chlorella essential oil and fatty acids have inhibitory effect on proliferation of colon cancer cells and apoptosis-inducing effect, which is a potential anti-colon cancer drug.
