目的:探讨IFN-γ对免疫编辑后人肺癌A549细胞对CIK杀伤敏感性的逆转作用。方法:应用RT-PCR和MTT方法分别检测IFN-γ对免疫编辑后A549细胞表面MICA表达的影响及A549细胞对CIK杀伤敏感性的变化。结果:免疫编辑后A549细胞表面低表达MICA,对CIK细胞杀伤敏感性低。IFN-γ可明显上调编辑后A549细胞MICA mRNA的表达率,提高CIK对编辑后A549细胞的细胞毒活性。结论:IFN-γ可以增强A549细胞表面MICA mRNA表达,逆转免疫编辑后A549细胞对CIK细胞杀伤敏感性。
Objective: To investigate the reversal effect of IFN-γ on CIK killing sensitivity of human lung cancer A549 cells after immuno-editing. Methods: The effects of IFN-γ on the MICA expression on A549 cells and the sensitivity to A549 cells to CIK were detected by RT-PCR and MTT respectively. Results: Immunofluorescence showed that the expression of MICA was low on the surface of A549 cells, and its sensitivity to CIK cells was low. IFN-γ significantly up-regulated the expression of MICA mRNA in A549 cells and increased the cytotoxic activity of CIK on A549 cells. CONCLUSION: IFN-γ can enhance the expression of MICA mRNA on A549 cells and reverse the sensitivity of A549 cells to CIK cell killing after immuno-editing.
