目的:观察葡萄籽原花青素对自发性高血压大鼠(SHR)左室重塑的保护作用并探讨其机制。方法:8周龄雄性SHR24只随机分成4组,每组6只:模型组、葡萄籽原花青素50mg/kg剂量组和200mg/kg剂量组、卡托普利组(30mg/kg)。6只同龄雄性Wistar-Kyoto大鼠设为对照组。治疗8周后,测定尾动脉收缩压(SBP);观察大鼠左室结构,测量左室后壁厚度;Masson染色下观察心肌血管周围胶原纤维情况并测量心肌血管周围胶原面积(PVCA);生化法和ELISA法分别检测左室心肌组织中NO和Ang II含量;Western blot测定心肌组织中ET-1蛋白表达。结果:葡萄籽原花青素能显著降低SHR尾动脉SBP,减轻左室壁增厚,减少心肌组织胶原、Ang II含量和ET-1蛋白表达,增加NO含量。葡萄籽原花青素200mg/kg剂量组的作用尤为显著。结论:葡萄籽原花青素对SHR左室重塑有减轻作用,可能与葡萄籽原花青素降低尾动脉SBP、减少Ang II含量、ET-1蛋白表达和增加NO产生有关。
Objective: To observe the protective effect of grape seed proanthocyanidins on left ventricular remodeling in spontaneously hypertensive rats (SHR) and to explore its mechanism. Methods: Male 8-week-old SHR24 were randomly divided into 4 groups with 6 mice in each group: model group, grape seed proanthocyanidin 50mg / kg dose group and 200mg / kg dose group, captopril group (30mg / kg). Six male Wistar-Kyoto rats of the same age were set as a control group. After 8 weeks of treatment, the tail artery systolic pressure (SBP) was measured. The left ventricular structure was observed and the thickness of the posterior wall of the left ventricle was observed. Masson staining was used to observe the collagen fibers in myocardium and to measure the area of perivascular collagen (PVCA) The contents of NO and Ang II in left ventricular myocardium were detected by ELISA and ELISA respectively. The protein expression of ET-1 in myocardium was detected by Western blot. RESULTS: Grape seed proanthocyanidins could significantly reduce SBP of SHR tail artery, relieve left ventricular wall thickening, decrease myocardial collagen, Ang II content and ET-1 protein expression and increase NO content. Grape seed proanthocyanidin 200mg / kg dose group is particularly significant role. CONCLUSION: Grape seed proanthocyanidins can reduce the left ventricular remodeling of SHR, which may be related to the reduction of SBP, the decrease of Ang II content, the expression of ET-1 and the increase of NO in grape seed proanthocyanidins.
