血栓素A_2(TxA_2)在血小板聚集中起重要作用,可引起冠状动脉血栓形成,导致缺血性心脏病(IHD)。细胞内游离钙是血小板聚集的主要介体。作者研究了 IHD 病人与健康人对 TxA_2引起的血小板聚集及细胞内Ca~(2+)浓度的变化,观察 IHD 病人血小板是否对 TxA_2更为敏感,及其与 Ca~(2+)移动的关系。因 TxA_2不稳定,故采用稳定、并具有同样生物活性的同类物 STA_2(9,11-ep-thio-11,12 methano-TxA_2)代替 TxA_2。
Thromboxane A 2 (TxA 2) plays an important role in platelet aggregation and can cause coronary thrombosis, leading to ischemic heart disease (IHD). Intracellular free calcium is the main mediator of platelet aggregation. The authors studied platelet aggregation and intracellular Ca 2+ concentrations induced by TxA 2 in IHD patients and healthy individuals and determined whether platelets in the IHD patients are more susceptible to TxA 2 and their relationship to Ca 2+ mobilization . Due to the instability of TxA_2, STA_2 (9,11-ep-thio-11,12 methano-TxA_2), which is stable and has the same bioactivity, was used instead of TxA_2.
