目的 :研究扶正祛邪丹对 K562细胞株和小鼠造血干细胞的影响 ,探讨扶正祛邪丹治疗骨髓增生异常综合征的疗效机理。方法 :扶正祛邪丹含药兔血清培养 K562细胞株 ,用流式细胞仪检测 P53蛋白表达、细胞活力、细胞凋亡与细胞周期 ;用造血干细胞培养法观察扶正祛邪丹对 BALB/C小鼠骨髓造血干细胞增殖的影响。结果 :扶正祛邪丹对 K562细胞株 P53蛋白表达有抑制作用 ,对红系造血祖细胞有促进作用。不能诱导细胞凋亡 ,对细胞增殖周期无明显影响 ,对小鼠骨髓粒系造血干细胞无明显促进作用。结论 :扶正祛邪丹促进红系造血干细胞增殖 ,降低 P53蛋白表达 ,可能为其主要疗效机理。
Objective : To study the effect of Fuzheng Qinxiedan on K562 cell line and mouse hematopoietic stem cells, and explore the therapeutic mechanism of Fuzhengxiexiedan in the treatment of myelodysplastic syndrome. Methods: The K562 cell line was cultured with Fuzheng Decoction containing rabbit serum. The expression of P53 protein, cell viability, cell apoptosis and cell cycle were detected by flow cytometry. The hematopoietic stem cell culture method was used to observe the effects of Fuzhengqiedan on BALB/C. Effect of murine bone marrow hematopoietic stem cell proliferation. RESULTS : The Fuzheng Decoction had inhibitory effect on the expression of P53 protein in K562 cell line and it promoted the erythroid hematopoietic progenitor cells. Can not induce apoptosis, no significant effect on the cell proliferation cycle, no significant role in the promotion of mouse bone marrow hematopoietic stem cells. Conclusion : Fuzheng Qinxie promotes the proliferation of erythroid hematopoietic stem cells and reduces the expression of P53 protein, which may be the main therapeutic mechanism.
