目的:观察山楂总黄酮联合替米沙坦对自发性高血压病大鼠血浆黏附分子的影响,并探讨其临床意义及可能的机制。方法30只自发性高血压大鼠随机分为对照组、替米沙坦组和联合给药组(山楂黄酮+替米沙坦组)各10只。对照组予以蒸馏水2 mL/d灌胃;替米沙坦组予以替米沙坦8 mg?kg-1?d-1灌胃;联合给药组予以替米沙坦8 mg?kg-1?d-1+山楂黄酮60 mg?kg-1?d-1灌胃。每隔2周测定大鼠尾动脉血压,取其平均值;12周末取大鼠腹主动脉肝素抗凝血3 mL,测定各组大鼠血浆细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)、一氧化氮(NO)、超氧化物歧化酶(SOD)和丙二醛(MDA)变化。结果与对照组比较,12周末替米沙坦组和联合给药组大鼠收缩压明显降低(P<0.01),而替米沙坦组和联合给药组间差异无统计学意义(P>0.05);与对照组比较,12周末替米沙坦组和联合给药组大鼠血浆ICAM-1、VCAM-1和MDA含量明显降低(P<0.01),而与替米沙坦组比较,联合给药组血浆ICAM-1、VCAM-1和MDA含量显著降低,差异有统计学意义(P<0.05);12周末,替米沙坦组和联合给药组大鼠血浆NO和SOD水平较对照组明显升高(P<0.01),而与替米沙坦组比较,联合给药组大鼠血浆NO和SOD水平升高,差异有统计学意义(P<0.05)。结论山楂总黄酮能有效地降低自发性高血压大鼠血浆黏附分子水平,升高NO含量,对血管内皮细胞起保护性作用。其机制可能与降低血浆MDA含量,升高SOD水平有关。“,”Objective To observe the effect of hawthorn leaffl avonoids (HLF) combined with telmisartan on plasma adhesion molecule in idiohypertensive rats and to investigate the clinical signifi cance and conceivable mechanism.Methods 30 idiohypertensive rats were divided into 3 groups randomly: 10 rats in control group, 10 rats in telmisartan group and 10 rats in HLF + telmisartan group. Control group were given intragastric administration with distilled water, telmisartan group were given with telmisartan (8 mg?kg-1?d-1),while combined group with telmisartan (8 mg?kg-1?d-1) + HLF (60 mg?kg-1?d-1). The rats caudal artery pressure were measured once two weeks. At the end of 12 weeks, serum levels of ICAM-1, VCAM-1, NO, SOD and MDA were serially assessed.Results Compared with control group, systolic pressure of rats in telmisartan group and combined group were depressed obviously at the end of 12 weeks (P<0.01). Meanwhile no difference was found between two treatment groups. Serum levels of ICAM-1, VCAM-1 and MDA in two treatment groups decreased signifi cantly comparing with control group after 12 weeks (P<0.01). However, compared with telmisartan group, serum levels of ICAM-1, VCAM-1 and MDA in combined group decreased signifi cantly (P<0.05). At the end of 12 weeks, serum levels of NO and SOD in two treatment groups increased signifi cantly (P<0.01), and combined group were more apparently (P<0.05).Conclusion HLF can depress the level of plasma adhesion molecule in idiohypertensive rats, and increase NO effectively. So that, it has benefi cial effects on endothelial function. The conceivable mechanism is the effect on decreasing MDA and increasing SOD.
