人白细胞介素3(IL-3)作用于早期造血并具有广谱效应。为了制备足量高纯度的IL-3,本研究利用分子遗传学技术构建了三株重组IL-3表达菌株,其中一株表达水平为16%。菌体裂解物对人粒-巨噬系祖细胞有刺激增生的作用;对小鼠早期与晚期红系祖细胞亦有刺激作用,对晚期的作用强;但是对小鼠粒-巨噬系祖细胞无刺激作用,提示人IL-3的造血刺激作用可能具有种属特异性。
Human Interleukin 3 (IL-3) acts on early hematopoiesis and has a broad spectrum of effects. In order to prepare a sufficient amount of high purity IL-3, three recombinant IL-3-expressing strains were constructed by using molecular genetics technique, in which the expression level of one strain was 16%. Cell lysate on human granulocyte macrophage progenitor cells stimulate the proliferation of the role of early and late mouse erythroid progenitor cells also stimulate the role of the late strong; Cells without stimulation, suggesting that human IL-3 hematopoietic stimulation may have species-specific.
