目的研究黄荆子Vitexnegundo抗肿瘤的有效部位及化学成分。方法采用MTT法进行体外抗肿瘤活性筛选;采用裸鼠移植瘤实验进行体内抗肿瘤活性验证;采用多种色谱技术进行分离纯化,经理化性质及波谱数据分析进行结构鉴定。结果从有效部位中分离并鉴定了8个化合物:对羟基苯甲酸(Ⅰ)、3,4-二羟基苯甲酸(Ⅱ)、芥子醛(Ⅲ)、阿魏醛(Ⅳ)、丁香酸(Ⅴ)、6-羟基-4-(4-羟基-3-甲氧基苯基)-3-羟甲基-7-甲氧基-3,4-二氢-(3R,4S)-2-醛基萘(Ⅵ)、6-羟基-4-(4-羟基-3-甲氧基苯基)-3-羟甲基-5-甲氧基-3,4-二氢-(3R,4S)-2-醛基萘(Ⅶ)、3-甲氧基-4-羟基苯甲酸(Ⅷ)。结论黄荆子醋酸乙酯萃取部位为抗肿瘤的有效部位,化合物Ⅲ、Ⅳ为首次从该属植物中分离得到。
Objective To study the effective anti-tumor and chemical components of Vitexnegundo. Methods The MTT assay was used to screen the anti-tumor activity in vitro. The tumor xenografts in nude mice were used to verify the anti-tumor activity in vivo. The chromatographic techniques were used to separate and purify the samples. As a result, eight compounds were isolated and identified from the effective fraction: p-hydroxybenzoic acid (I), 3,4-dihydroxybenzoic acid (II), sibutamide (III), ferulaldehyde (IV), syringic acid (V) ), 6-Hydroxy-4-(4-hydroxy-3-methoxyphenyl)-3-hydroxymethyl-7-methoxy-3,4-dihydro-(3R,4S)-2-aldehyde Vinylnaphthalene (VI), 6-hydroxy-4-(4-hydroxy-3-methoxyphenyl)-3-hydroxymethyl-5-methoxy-3,4-dihydro-(3R,4S) 2-Aldehyde naphthalene (VII), 3-methoxy-4-hydroxybenzoic acid (VIII). Conclusion The ethyl acetate extracted from Vitex negundo L. is an effective antitumor site. Compounds III and IV are isolated from this genus for the first time.
