目的在辽宁省采集蚊虫标本进行病毒分离,了解虫媒病毒分布情况。方法2008年夏季在辽宁省丹东和锦州市采集蚊虫标本,利用C6/36和BHK-21等细胞分离病毒;对新分离病毒利用血清学、分子生物学和生物信息学方法进行鉴定。结果在辽宁省丹东和锦州市共采集蚊虫标本3属5种9296只,包括库蚊属的三带喙库蚊、淡色库蚊,伊蚊属的刺扰伊蚊、背点伊蚊和按蚊属的中华按蚊。从蚊虫标本中共得到4株病毒分离物,其中1株为乙型脑炎(乙脑)病毒(丹东市的三带喙库蚊),2株为版纳病毒(锦州市的中华按蚊),1株为辽宁病毒和版纳病毒混合(锦州市的中华按蚊)。新分离乙脑病毒属于基因Ⅰ型乙脑病毒,E基因编码病毒毒力和抗原表位的位点未发生改变。结论在辽宁省再次分离到乙脑病毒,并首次证明辽宁省存在辽宁病毒。
Objective To collect mosquito specimens in Liaoning Province for virus isolation and to understand the distribution of arboviruses. Methods In summer 2008, mosquitoes were collected from Dandong and Jinzhou in Liaoning Province. Viruses were isolated from C6 / 36 and BHK-21 cells. The newly isolated viruses were identified by serological, molecular biology and bioinformatics methods. Results A total of 9296 mosquitoes were collected from 3 genera and 5 species of mosquitoes in Dandong and Jinzhou of Liaoning Province, including Culex tritaeniorhynchus, Culex pipiens pallens, Culex pipiens pallens, Aedes albopictus and Aedes albopictus Anopheles sinensis. A total of 4 virus isolates were obtained from mosquito specimens, of which 1 strain was JE virus (Culex tritaeniorhynchus) in Dandong, 2 were BAV (Anopheles sinensis) in Jinzhou, 1 The strain was a mixture of Liaoning virus and Banna virus (Anopheles sinensis in Jinzhou). The newly isolated JE virus belongs to the genotype I JEV, and the site of the E gene encoding the virulence and epitopes has not changed. Conclusion The JE virus was isolated again in Liaoning Province and Liaoning Province was the first to be confirmed.
