目的观察雷公藤红素对人类急性髓细胞白血病(acute myeloid leukemia,AML)细胞增殖和凋亡的影响。方法初诊人AML细胞按雷公藤红素浓度分成5组,A组为阴性对照组,B~E组分别为雷公藤红素1,2,5,10μmol/L处理组,应用四甲基偶氮唑兰比色法及流式细胞术等方法检测雷公藤红素对细胞的生长增殖抑制作用及诱导凋亡作用。结果作用不同时间后,B~E组增殖抑制率随雷公藤红素浓度的增加而增加,与A组比较差异均有统计学意义(P<0.01);C,D组增殖抑制率与B组比较差异有统计学意义(P<0.01),与E组比较差异无统计学意义(P>0.05);雷公藤红素抑制作用的最适浓度为2μmol/L;C~E组作用6,12,24h时增殖抑制率比较差异无统计学意义(P>0.05),与3h时比较差异均有统计意义(P<0.01);B~E组作用于AML细胞6h后均能诱导AML细胞凋亡,D组细胞凋亡率最高;2μmol/L雷公藤红素作用于AML细胞3,6,12,24h,其细胞凋亡率随时间延长而逐渐增高。结论雷公藤红素有明确抗AML细胞增殖的能力,其作用可能与诱导细胞凋亡有关。
Objective To observe the effect of tripterine on the proliferation and apoptosis of human acute myeloid leukemia (AML) cells. The apoptosis of AML cells induced by B ~ E group was significantly higher than that of AML cells at 6h (P> 0.05), and the difference was not statistically significant , And the apoptosis rate in group D was the highest. The apoptosis rate of AML cells treated with 2μmol / L tripterine for 3,6,12,24h increased gradually with time. Conclusion Tripterine has a clear ability of anti-proliferation of AML cells, which may be related to the induction of apoptosis.
