目的建立实时荧光定量逆转录聚合酶链式反应(real-time RT PCR)法对手足口病病例标本进行柯萨奇病毒A6(CoxA6)病原检测,明确病原,为手足口病的防治对策提供依据。方法用real-time RT-PCR法对2015年5月-6月份手足口病标本进行EV(总肠道病毒)、EV71、CoxA16核酸检测,对其他肠道病毒(非EV71和非CoxA6)阳性标本再进行CoxA6核酸检测,选取CoxA6核酸1份阴性和3份阳性提取物,送河南省疾病预防控制中心复核验证检测的准确性。结果 167份标本中EV71阳性17份,CoxA16阳性13份,其他肠道病毒阳性61份;4份CoxA6核酸省级验证结果一致。结论 CoxA6是濮阳市手足口病的又一主要病原,在普通和重症病例中都存在,real-time RT-PCR法用于CoxA6检测能为临床诊断和疾病预防控制提供可靠数据。
Objective To establish a real-time RT-PCR method for the detection of Coxsackievirus A6 (CoxA6) pathogen in hand-foot-mouth disease patients and to clarify the pathogen and provide a basis for prevention and treatment of hand-foot-mouth disease . Methods The EV (total enteroviruses), EV71 and CoxA16 were detected by real-time RT-PCR in May-June 2015. The positive samples of other enterovirus (non-EV71 and non-CoxA6) CoxA6 nucleic acid test was carried out again. One negative and three positive CoxA6 nucleic acid extracts were selected and sent to Henan Provincial Center for Disease Control and Prevention for review and verification. Results Among the 167 samples, 17 were positive for EV71, 13 were positive for CoxA16 and 61 were positive for other enteroviruses. The results of 4 provincial CoxA6 nucleic acid tests were consistent. Conclusion CoxA6 is another major pathogen of hand-foot-mouth disease in Puyang City, and exists in both common and severe cases. The real-time RT-PCR assay for CoxA6 can provide reliable data for clinical diagnosis and disease prevention and control.
