目的:探讨stathmin基因表达水平与紫杉醇联合铂类疗效在卵巢癌中相关性研究及其临床意义。方法:应用Quanti-Gene Assay技术检测88例III~IV上皮性卵巢肿癌患者肿瘤组织中stathmin基因的表达水平,对照分析其与患者年龄、新辅助化疗,残瘤灶,药物敏感度、总生存期/生存率、stathmin蛋白表达的关系。结果:患者年龄越大,肿瘤组织中stathmin基因表达水平越高,但P>0.05,无统计学意义;新辅助化疗后,患者stathmin基因表达量增加,且有显著性差异,P<0.01;术后有残瘤灶患者的stathmin基因表达量较高,而没有残瘤的基因表达量较低,P<0.05,有统计学意义;药物敏感度较低患者的stathmin基因表达量较大,相反,敏感度较大的表达量较小,但通过单因素方差分析发现差异没有达到显著的程度;生存期越长,患者的stathmin基因表达量越低,两者呈负相关性,且有显著意义;OS曲线结果显示stathmin低表达组的生存期明高于stathmin高表达组;stathmin蛋白质强度越大,其RNA表达量就越高,其RNA表达与蛋白质的表达一致,有显著性差异。结论:肿瘤组织中stathmin基因表达水平可能与卵巢癌患者紫杉醇联合铂类耐药及预后密切相关。
Objective: To investigate the correlation between the expression of stathmin gene and the efficacy of paclitaxel and platinum in ovarian cancer and its clinical significance. Methods: Quanti-Gene Assay was used to detect the expression of stathmin in 88 cases of epithelial ovarian cancer with epithelial ovarian neoplasm. The control group was analyzed with age, neoadjuvant chemotherapy, residual tumor, drug sensitivity, total survival Period / survival rate, stathmin protein expression. Results: The stathmin gene expression level in tumor tissue increased with age, but there was no significant difference between two groups (P> 0.05). After neoadjuvant chemotherapy, the expression of stathmin gene increased in patients with significant difference (P <0.01) Stathmin gene expression was higher in patients with residual tumor, but the gene expression without residual tumor was lower (P <0.05, statistically significant). In patients with lower drug sensitivity, the expression of stathmin gene was higher, However, the single factor analysis of variance showed that the difference did not reach significant level. The longer the survival time was, the lower the stathmin expression in patients was, the two were negatively correlated and had significant significance. The OS curve showed that the survival of stathmin low expression group was higher than that of stathmin high expression group. The higher the stathmin protein intensity was, the higher the RNA expression was, and the expression of RNA was the same as that of protein. Conclusion: The expression of stathmin in tumor tissue may be closely related to the paclitaxel resistance and prognosis of patients with ovarian cancer.
