目的:探讨CD44v9 mRNA和MMP-2在PSTT中的表达及相关性。方法:应用组织芯片分别结合原位杂交和免疫组化方法检测PSTT中CD44v9 mRNA和MMP-2的表达情况。结果:CD44v9 mRNA在正常绒毛,良性PSTT,恶性PSTT的阳性表达率分别是:16.7%,30.3%,100%。其中恶性PSTT与正常绒毛,良性PSTT相比差异有统计学意义(P<0.05);而MMP-2的阳性表达率分别为:6.6%,30.7%,100%,其中恶性PSTT与正常绒毛、良性PSTT相比差异有统计学意义(P<0.05)。结论:CD44v9 mRNA和MMP-2的过表达促进PSTT的浸润、转移,二者具有正协同作用,联合检测CD44v9 mRNA和MMP-2蛋白可作为PSTT的浸润、转移及评价预后的生物学指标。
Objective: To investigate the expression and correlation of CD44v9 mRNA and MMP-2 in PSTT. Methods: The expressions of CD44v9 mRNA and MMP-2 in PSTT were detected by tissue microarray combined with in situ hybridization and immunohistochemistry respectively. Results: The positive rates of CD44v9 mRNA in normal villi, benign PSTT and malignant PSTT were 16.7%, 30.3% and 100% respectively. The positive rates of MMP-2 were 6.6%, 30.7% and 100% respectively in malignant PSTT compared with normal villus and benign PSTT (P <0.05) PSTT difference was statistically significant (P <0.05). Conclusion: Overexpression of CD44v9 mRNA and MMP-2 promote the invasion and metastasis of PSTT. Both of them have positive synergistic effect. Combined detection of CD44v9 mRNA and MMP-2 protein can be used as biological indicators of infiltration, metastasis and prognosis of PSTT.
