目的:探讨白藜芦醇对UVA照射后HaCaT细胞的光保护作用及其可能机制。方法:采用5J/cm2UVA照射HaCaT细胞后,分别加入0.01mmol/L和0.1mmol/L的白藜芦醇进行干预,同时设正常对照组与UVA照射组,分别用MTT法检测细胞增殖能力,用羟胺法、比色法、TBA法检测细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,电镜观察细胞超微结构的改变。结果:白藜芦醇能提高UVA照射后HaCaT细胞增殖能力、细胞SOD和GSH-Px活性,降低MDA含量,且呈剂量依赖性增加或降低(P<0.05)。电镜结果显示白藜芦醇能减轻UVA对HaCaT细胞超微结构的损伤。结论:白藜芦醇能减轻UVA对HaCaT细胞增殖的抑制作用、超微结构和氧化功能的损伤,并有随剂量依赖性增加的保护作用,其机制可能与白藜芦醇提高氧化酶活性、清除自由基有关。
Objective: To investigate the photoprotective effect of resveratrol on UVA irradiated HaCaT cells and its possible mechanism. METHODS: After irradiated with 5J/cm2 UVA, HaCaT cells were treated with 0.01mmol/L and 0.1mmol/L resveratrol, respectively. The normal control group and UVA irradiation group were set up. The cell proliferation was detected by MTT assay. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) were measured by hydroxylamine method, colorimetric method and TBA method. The ultrastructure of cells was observed by electron microscope. change. RESULTS: Resveratrol could increase the proliferation of HaCaT cells, the activity of SOD and GSH-Px, and decrease the content of MDA in UVA-irradiated cells, and increased or decreased in a dose-dependent manner (P<0.05). The results of electron microscopy showed that resveratrol could reduce the ultrastructure damage of HaCaT cells by UVA. CONCLUSION: Resveratrol can reduce the inhibitory effect of UVA on the proliferation of HaCaT cells, damage the ultrastructure and oxidative function, and has a protective effect in a dose-dependent manner.
