应用胆固醇生物合成的体外酶反应筛选系统,从1000多株真菌中筛选出具有抑制胆固醇生物合成活性的SIPI-8926菌株;对SIPI-8926菌株代谢产物研究,应用溶媒萃取、硅胶柱分离、LH20凝胶层析和重结晶等方法,从其发酵液中分离出活性成分SIPI-8926-Ⅳ。通过元素分析和质谱,确定其分子式为:C30H50O3(分子量为:458.376),综合紫外光谱、红外光谱、1H-NMR和13C-NMR等图谱数据的解析,确定SIPI-8926-Ⅳ为三萜类化合物,结构与中药来源的大豆黄醇B(soyasapogenolB)一致,化学命名为:3β,21β,24-三羟基-齐果墩烷-12-烯
In vitro enzyme reaction screening system of cholesterol biosynthesis, SIPI-8926 strain with inhibitory activity of cholesterol biosynthesis was screened from more than 1000 strains of fungi. The metabolites of SIPI-8926 strain were studied by solvent extraction, silica gel column, LH20 condensate Gel chromatography and recrystallization, the active ingredient SIPI-8926-IV was isolated from the fermentation broth. The molecular formula of C30H50O3 (molecular weight: 458.376) was determined by elemental analysis and mass spectrometry.
