目的:研究白簕叶总黄酮的大孔树脂纯化工艺。方法:通过静态、动态吸附从8种大孔树脂中确定白簕叶总黄酮精制最优树脂为HPD-600,以黄酮吸附率、洗脱率、黄酮纯度和薄层色谱结果为评价指标,综合评判纯化最优工艺。结果:其最佳吸附工艺参数为:上样量30mL,上样浓度4mg·mL-1、上样液pH5~6、吸附流速0.5mL·min-1;最佳洗脱条件为:洗脱液体积40mL,pH5~6,70%的乙醇溶液,洗脱流速2mL·min-1,精制后黄酮质量分数超过50%。TLC结果显示,不同浓度的洗脱液中均含有芦丁和金丝桃苷,pH1~2、3~4、5~6的洗脱液均含有芦丁和金丝桃苷。结论:采用HPD-600树脂精制白簕叶总黄酮,操作简单,成本低廉,精制效果突出,有较高的工业生产应用价值。
Objective: To study the macroporous resin purification technology of total flavones of Methods: The optimum resin of total flavonoids from buckwheat leaf was identified as HPD-600 by static and dynamic adsorption. The flavonoid adsorption rate, elution rate, flavonoid purity and TLC were used as evaluation indexes, Judge the best purification process. Results: The optimal adsorption parameters were as follows: sample volume 30mL, loading concentration 4mg · mL-1, pH5 ~ 6 and flow rate 0.5mL · min-1. The optimum elution conditions were as follows: Volume 40mL, pH5 ~ 6,70% ethanol solution, the elution flow rate 2mL · min-1, flavonoids after purification mass fraction of more than 50%. The results of TLC showed that rutin and hyperoside were contained in different concentrations of eluent, rutin and hyperoside contained in eluent of pH1 ~ 2,3 ~ 4,5 ~ 6 respectively. Conclusion: Using HPD-600 resin to purify the total flavonoids of the leaves, the operation is simple, the cost is low, the refining effect is outstanding, and has high industrial application value.
