目的观察酸甘生津方及其拆方对干燥综合征(SS)模型小鼠血清IgG含量及颌下腺BAFF表达的影响。方法将60只BALB/C小鼠随机分为全方组、养阴组、活血组、解毒组、模型组及空白组,除空白组外其余各组小鼠均采用同种小鼠颌下腺接种方法造模。造模第3周起,分别予以酸甘生津方全方及其拆方制剂灌胃,模型组及空白组给予等量蒸馏水灌胃。干预2周后观察各组小鼠血清IgG含量及颌下腺组织BAFF表达水平。结果与空白组比较,模型组小鼠血清IgG含量、BAFF表达水平均升高,差异有统计学意义(P<0.01)。与模型组比较,各药物组小鼠血清IgG含量、BAFF表达水平均降低,差异有统计学意义(P<0.01)。各药物组之间比较,小鼠血清IgG含量差异无统计学意义(P>0.05)。与全方组比较,3个拆方组BAFF表达水平差异有统计学意义(P<0.01);与解毒组比较,养阴组与活血组BAFF表达差异有统计学意义(P<0.01);养阴组与活血组之间比较,BAFF表达差异无统计学意义(P>0.05)。结论酸甘生津方及其拆方均能有效降低SS疾病模型小鼠血清IgG含量,显著抑制SS模型小鼠颌下腺组织BAFF过度表达,全方组效果最佳,这可能是其有效治疗SS的作用机制之一。
Objective To observe the effects of sankangshenjinfang and its decomposed formulas on the serum IgG level and the expression of BAFF in submandibular glands of mice with Sjogrens syndrome (SS). Methods Sixty BALB / C mice were randomly divided into the whole group, Yangyin group, Huoxue group, detoxification group, model group and blank group. Except for the blank group, the mice in other groups were all inoculated subcutaneously Modeling. From the third week of model making, Ganshen Shengjinfang and its disassembled preparations were given respectively, and the model group and the blank group were given the same amount of distilled water. After 2 weeks’ intervention, the serum IgG levels and the expression of BAFF in submandibular gland were observed. Results Compared with the blank group, the levels of serum IgG and the expression of BAFF in model group were significantly increased (P <0.01). Compared with the model group, the serum IgG levels and the expression of BAFF in each drug group decreased, with statistical significance (P <0.01). There was no significant difference in serum IgG between the two groups (P> 0.05). The BAFF expression levels in three groups were statistically significant (P <0.01). Compared with the detoxification group, there was significant difference in BAFF expression between Yangyin group and Huoxue group (P <0.01) There was no significant difference in BAFF expression between the negative group and the active blood group (P> 0.05). Conclusion SGD and its disassembled prescriptions can both effectively reduce the serum IgG level in SS model mice and significantly inhibit BAFF overexpression in submandibular gland tissue of SS model mice, which may be the best way to treat SS One of the mechanisms.
