目的:研究千金子素L1、千金子素L2、千金子素L8、6(17),12(E)-千金二萜醇-5,15-二乙酸-3-苯乙酸酯在Caco-2细胞中的代谢产物及途径。方法:100μg/mol千金子二萜类化合物作用于Caco-2细胞3、6、12 h后收集并纯化样品,LC/MS/MS分析鉴定代谢产物。结果:千金子素L1在Caco-2细胞中首先发生酯基水解,然后再被甲基化,最终鉴定得到两个甲基化产物;千金子素L2、千金子素L8及6(17),12(E)-千金二萜醇-5,15-二乙酸-3-苯乙酸酯各鉴定出一个酯基水解产物。结论:千金子素L1在Caco-2细胞中主要代谢途径为酯基水解和甲基化;千金子素L2、千金子素L8及6(17),12(E)-千金二萜醇-5,15-二乙酸-3-苯乙酸酯在Caco-2细胞中主要代谢途径为酯基水解。LC/MS/MS可以快速、灵敏地鉴定千金子素L1、千金子素L2、千金子素L8及6(17),12(E)-千金二萜醇-5,15-二乙酸-3-苯乙酸酯在Caco-2细胞中代谢产物。
OBJECTIVE: To investigate the effect of cadmium on lipoprotein L1, sequon L2, sequins L8,6 (17), 12 (E) -dimeridene-5,15-diacetate-3-phenylacetate in Caco-2 Metabolites and pathways in cells. METHODS: 100 μg / mol dione was isolated and purified from Caco-2 cells after 3, 6 and 12 h. The metabolites were identified by LC / MS / MS. Results: Chlortetracycline L1 was first esterified in Caco-2 cells and then methylated, and finally two methylated products were obtained. Chlortetracycline L2, cadmium L8 and 6 (17), 12 (E) -dimerpeneol-5,15-diacetic acid-3-phenylacetate each identified one ester hydrolysate. CONCLUSION: The main metabolic pathways of connexin L1 in Caco-2 cells are ester-based hydrolysis and methylation; sequon L2, sequins L8 and 6 (17), 12 (E) -dieno ol-5 The main metabolic pathway of 15-diacetic acid-3-phenylacetate in Caco-2 cells is ester hydrolysis. LC / MS / MS provides a rapid and sensitive identification of the fametin L1, the ficin L2, the ficin L8 and 6 (17), the 12 (E) -dimerpene-5,15-diacetic acid-3- Phenylacetate metabolites in Caco-2 cells.
