目的:探讨大鼠海马CA1区微注射L-精氨酸(L-Arg)对大鼠空间学习记忆的影响。进而了解L-Arg的作用和机理,为深入研究该药物的神经生物学机制提供实验依据。方法:10只SD大鼠随机分为实验组(n=5,微量注射L-Arg 0.5μl,浓度为50g/L)和对照组(n=5,微量注射生理盐水0.5μl),利用八臂迷宫对大鼠进行测试,记录不同药物注射下大鼠在八臂迷宫中的工作记忆错误和参考记忆错误。结果:大鼠海马区微注射生理盐水前后八臂迷宫实验工作记忆错误分别为2±0.3次、2.4±0.2次,前后无显著变化(P>0.05);注药前后参考记忆错误分别为6.2±0.3次、6.4±0.2次,前后无显著变化(P>0.05)。实验测量结果对比发现,导管植入手术对大鼠空间学习记忆能力无明显影响;大鼠海马区微注射L-Arg前后测试时间分别为224.1s、223.8s,注射前后无明显差异(P>0.05)。大鼠海马区注射L-Arg后工作记忆错误为1.9±0.3次,对照组为2.7±0.4次,实验组工作记忆错误次数明显降低(P<0.05);注药后参考记忆错误分别为实验组5.3±0.4次、对照组6.5±0.5次,实验组参考记忆错误次数明显降低(P<0.05)。结论:海马区微注射生理盐水对大鼠的学习记忆能力影响不大,海马区微注射L-Arg能提高大鼠的学习记忆能力。
Objective: To investigate the effect of microinjection of L-arginine (L-arginine) into hippocampal CA1 area on spatial learning and memory in rats. And then understand the role and mechanism of L-Arg, to provide experimental evidence for further study of the neurobiological mechanism of the drug. Methods: Ten SD rats were randomly divided into experimental group (n = 5, 0.5μl L-Arg, concentration 50g / L) and control group (n = 5, 0.5μl microinjection of saline) The maze was used to test the rats, recording the working memory errors and reference memory errors of the rats in the eight-armed maze under different drug injections. Results: There were no significant changes (P> 0.05) in the memory error of the 8-arm maze before and after microinjection of saline into the hippocampus of rats. The errors of the reference memory before and after injection were 6.2 ± 0.3 times, 6.4 ± 0.2 times, no significant changes before and after (P> 0.05). The comparison of the experimental results showed that catheterization had no significant effect on spatial learning and memory in rats. The time before and after microinjection of L-Arg in hippocampus of rat was 224.1s and 223.8s respectively, with no significant difference before and after injection (P> 0.05 ). The working memory error was 1.9 ± 0.3 times in the hippocampus of rats after injection of L-Arg, 2.7 ± 0.4 times in the control group, and the number of working memory errors in the experimental group was significantly decreased (P <0.05). The reference memory errors were the experimental group 5.3 ± 0.4 times, 6.5 ± 0.5 times in the control group. The number of reference memory errors in the experimental group was significantly decreased (P <0.05). Conclusion: Microinjection of normal saline into hippocampus has little effect on learning and memory in rats. Microinjection of L-Arg into hippocampus can improve learning and memory in rats.
