目的建立超高效液相色谱-荧光法(UPLC-FLD)测定婴幼儿乳粉中胆碱含量的方法。方法利用1-萘基异氰酸酯与胆碱反应生成稳定的、具有荧光的阳离子芳香族化合物,用超高效液相色谱-荧光法分离测定。乳粉样品经盐酸水解,无水硫酸钠脱水后与1-萘基异氰酸酯进行衍生反应,经ACQUITY UPLC BEH HILIC色谱柱分离,用流动相四甲基氢氧化铵-羟基乙酸-乙腈-水溶液进行洗脱,然后通过荧光检测器(FLD)进行检测,激发波长和发射波长分别为220 nm和350 nm。结果胆碱的浓度(以胆碱阳离子计)在0.90μg/ml~13.5μg/ml时线性关系良好(相关系数r=0.999 7),回收率为89.0%~92.5%,仪器的检出限为0.052μg/ml。结论该方法具有成本低、灵敏度高、快速、简便等优点。
Objective To establish a method for the determination of choline in infant milk powder by ultra performance liquid chromatography-fluorescence (UPLC-FLD). The method utilizes the reaction of 1-naphthyl isocyanate and choline to generate stable, fluorescent cationic aromatic compounds and their separation by ultra performance liquid chromatography-fluorescence spectrometry. The milk powder samples were hydrolyzed by hydrochloric acid, dehydrated with anhydrous sodium sulfate and then subjected to derivatization with 1-naphthyl isocyanate, separated on an ACQUITY UPLC BEH HILIC column and washed with a mobile phase of tetramethylammonium hydroxide-glycolate-acetonitrile-water Off and then detected by Fluorescence Detector (FLD) with excitation and emission wavelengths of 220 nm and 350 nm, respectively. Results The linearity of choline (choline cation) was good at 0.90μg / ml ~ 13.5μg / ml (correlation coefficient r = 0.999 7) and the recovery was 89.0% ~ 92.5%. The detection limit was 0.052 μg / ml. Conclusion The method has the advantages of low cost, high sensitivity, fastness and convenience.
