目的:采用高效液相色谱-高分辨飞行时间质谱(HPLC-TOF/MS)对中药复方小柴胡汤中的化学成分进行快速鉴别。方法:色谱柱Agilent ZORBAXSB-C18(100mm×3.0mm,3.5μm),甲醇与0.1%的甲酸水溶液梯度洗脱,柱温为25℃,进样体积4μl,流速0.6ml/min,检测器为飞行时间质谱,ESI离子源,质量数扫描范围m/z200~1500。结果:一次性鉴别出小柴胡汤中38个化学成分。结论:采用HPLC-TOF/MS对小柴胡汤中的化学成分进行了色谱表征,一次性在一张图谱上共表征出38个化学成分,这可为中药复方小柴胡汤中化学成分的体内代谢及作用机制的深入研究奠定基础。
OBJECTIVE: To rapidly identify the chemical constituents of traditional Chinese medicine compound Xiao Chai Hu Tang by high performance liquid chromatography-high resolution liquid chromatography-time of flight mass spectrometry (HPLC-TOF / MS). Methods: The column was Agilent ZORBAXSB-C18 (100mm × 3.0mm, 3.5μm) with a gradient of methanol and 0.1% formic acid. The column temperature was 25 ℃. The injection volume was 4μl and the flow rate was 0.6ml / min. Time mass spectrometry, ESI ion source, mass scanning range m / z200 ~ 1500. Results: One-time identification of Xiao Chai Hu Tang 38 chemical composition. Conclusion: The chemical constituents in Xiao Chai Hu Tang were characterized by HPLC-TOF / MS. The 38 chemical constituents were identified in one map at a time, which could be the mechanism of the in vivo metabolism of Chinese herbal compound Xiao Chai Hu Tang In-depth study laid the foundation.
