目的探讨大肠癌中淋巴管密度(LVD)和血管内皮生长因子-A(VEGF-A)的表达与微血管密度(MVD)的关系。方法应用抗podoplanin及抗VEGF-A单克隆抗体检测大肠癌组织及相应正常大肠组织中的LVD,并计数CD34阳性着色的大肠癌组织MVD。结果①大肠癌组织中的LVD明显高于相应的正常组织(P<0.01),LVD与淋巴管内癌栓密切相关(P<0.05)。②VEGF-A在大肠癌组织中的表达显著高于相应正常组织(P<0.01),VEGF-A的表达与MVD(P<0.05)相关但与LVD无关(P>0.05)。③大肠癌中MVD显著高于LVD(P<0.01),但两者无关。结论淋巴管的形成在大肠癌的发生、发展过程中起重要的作用;VEGF-A在大肠癌组织中可诱导微血管形成但与淋巴管的形成无关;大肠癌的MVD显著高于LVD,但两者无相关性,提示血管形成及淋巴管形成在大肠癌的发生、发展过程中可能为相互独立的过程。
Objective To investigate the relationship between the expression of lymphatic vessel density (LVD) and vascular endothelial growth factor-A (VEGF-A) and the microvessel density (MVD) in colorectal cancer. Methods The anti-podoplanin and anti-VEGF-A monoclonal antibodies were used to detect LVD in colorectal cancer tissues and their corresponding normal colorectal tissues. MVD in CD34 positive colorectal cancer tissues was counted. Results ① The LVD in colorectal cancer tissues was significantly higher than that in corresponding normal tissues (P <0.01). The LVD was closely related to the tumor thrombus in lymphatic vessels (P <0.05). ② The expression of VEGF-A in colorectal cancer tissues was significantly higher than that in corresponding normal tissues (P <0.01). The expression of VEGF-A was correlated with MVD (P <0.05) but not with LVD (P> 0.05). ③ The MVD in colorectal cancer was significantly higher than that in LVD (P <0.01), but both had no correlation. Conclusions The formation of lymphatic vessels plays an important role in the carcinogenesis and development of colorectal cancer. VEGF-A can induce microvessel formation in colorectal cancer but has no relation with the formation of lymphatic vessels. The MVD of colorectal carcinoma is significantly higher than that of LVD, No correlation, suggesting that the formation of blood vessels and lymphangiogenesis in the occurrence and development of colorectal cancer may be independent process.
