目的:探讨自噬的调节对骨肉瘤化疗的影响。方法:建立裸鼠骨肉瘤动物模型,自噬抑制剂3甲基腺嘌呤(3MA)和顺铂(DDP)干预肿瘤的生长,观察不同药物对骨肉瘤的生长情况的影响,检测肿瘤大小变化,HE染色观察病理表现。结果:对照组、DDP组、3MA+DDP组3组之间用药前后体积的比较差异具有统计学意义。DDP确实能较有效的抑制肿瘤的生长,加入3MA干预后能增强其敏感性,肿瘤的生长得到了进一步的抑制。HE染色见3MA+DDP组移植瘤组织中明显的坏死区。结论:3MA联合DDP化疗能提高对骨肉瘤细胞的杀伤率。
Objective: To investigate the effect of autophagy regulation on the chemotherapy of osteosarcoma. Methods: The animal model of osteosarcoma in nude mice was established. The growth of osteosarcoma was observed with the intervention of 3MA and 3DP and the effect of different drugs on the growth of osteosarcoma. The changes of tumor size, HE staining for pathological findings. Results: There were significant differences in the volume of the three groups before and after treatment between the control group, the DDP group and the 3MA + DDP group. DDP can indeed inhibit the growth of tumor more effectively. The addition of 3MA can enhance the sensitivity of tumor growth and further inhibit the growth of tumor. Hematoxylin-eosin staining showed obvious necrosis in the xenografts of 3MA + DDP group. Conclusion: 3MA combined with DDP chemotherapy can increase the killing rate of osteosarcoma cells.
