目的优化中药东北天南星生物碱类成分提取、纯化工艺,确定最佳工艺参数。方法采用单因素考察结合正交实验设计,以胡芦巴碱含量、出膏率为指标筛选东北天南星生物碱类成分最佳提取工艺;采用大孔吸附树脂技术,通过静、动态吸附与解吸试验,优选最佳纯化工艺。结果中药东北天南星生物碱类成分最佳提取工艺为20倍量60%乙醇,回流提取2次,每次2h;最佳纯化工艺为HPD-300树脂,0.1g/ml浓度的提取液1∶1(药材∶干树脂)比例上样,5BV水洗脱除杂后,5BV 70%乙醇洗脱,以胡芦巴碱计,得到的天南星生物碱纯度提高了7倍。结论研究优化了东北天南星生物碱类成分的提取、纯化工艺,确定了最佳工艺参数,为其临床应用开发与工业化生产提供依据。
OBJECTIVE To optimize the extraction and purification of Aconitum alkaloids from the northeast of Chinese traditional medicine, and determine the optimum technological parameters. Methods The single factor test combined with orthogonal experimental design, with fenugreek alkali content, the rate of ointment as an index screening of the best extraction technology of Aconitum alkaloids in Northeast China; using macroporous resin technology, static and dynamic adsorption and desorption test , Preferably the best purification process. Results The optimum extraction technology of Anemarrhena asphodeloides was 20 times the amount of 60% ethanol and refluxed 2 times for 2 hours each time. The best purification technology was HPD-300 resin, the concentration of 0.1g / ml extract 1: 1 (Medicinal material: dry resin), the purity of Araceae alkaloid obtained was 7 times higher than that obtained after the 5BV water was eluted and removed by 5BV 70% ethanol. Conclusion The study optimized the extraction and purification of the components of Aconitum alkaloids in northeast China, and determined the best process parameters, which provided the basis for its clinical application development and industrial production.
